MEDIA CELL CULTURE - PAN BIOTECH

PAN-Biotech manufactures and supplies a wide range of high quality reagents for use in cell culture. To simplify the transformation process from FBS-dependent to FBS-free or xeno-free cell culture methods, and to effectively reduce the consumption of FBS in research and industry for more sustainability and reproducibility The absence of vitamins in culture may lead to decrease in cell growth, cell death or loss of productivity Cell culture media are not only a perfect breeding ground for your cell culture but also for many contaminants, such as bacteria and fungi. They are the main protein component in serum and are added to cell culture media to increase the stability of cell membranes and to bind possibly toxic elements. Cells are cultured in a medium containing 10% FBS of the tested batch. By flow cytometric analysis the cultured hMSCs are then assayed and analyzed for morphology, expansion and vitality of cells.

Attachment & Cryo

The gelatine solution is used for coating cell culture dishes. It is applied in adherent cell cultures working with e.g. endothelial cells or ES-cells. Fibronectin is a large glycoprotein widely distributed in soluble form in the plasma and body fluids. Many cell types synthesize fibronectin. There is also an insoluble form of fibronectin in tissues. Plasma fibronectin is not identical to cellular fibronectin but equally effective in supporting cell attachment. Fibronectin promotes the attachment and spreading of many adherent cells on plastic, but also mediates binding to other extracellular matrix components such as e.g. collagen.  Our freezing medium is recommended for the cryoconservation of cells. The medium is based on DMEM, supplemented with a mix of fetal bovine serum and DMSO. This composition guarantees a high survival rate and excellent cell growth after thawing.

Enzymes Cell Dissociation

Accutase® can be used as a direct replacement of Trypsin for cell dissociation. Accutase® is a ready-to-use cell detachment solution made of collagenolytic and proteolytic enzymes. It is used for routine detachment of adherent cells from tissue culture plates and flasks. A multitude of cell types has been successfully sub-cultured with Accutase®, including fibroblasts, endothelial, vascular smooth muscle cells, as well as hepatocytes, embryonal stem cells, and many immortalized cell lines such as adherent CHO and BHK cells, HEK 293, L929, HeLa, 3T3, and others. Serum Human

From collection to the final product  Human Sera is the future for human cell therapy.  Therefore the highly sensitive material and is subject to stringent safety EU regulations.  The sample collectionis taken from human, healthy donors in Europe, certified donations institutions.  The average age of our donors is approximately 26 years.  The collection and testing of human plasma is carried out in accordance with the current version of Good Manufacturing Practice Annex 14th.  Each donation is precisely tested regarding various biophysical parameters and viral infections and negative in the tests anit HIV 1/HIV 2, Hbs Ag, anti-HCV, HIV-1 Ag and/or HIV-1 NAT and Syphilis.  From the collection of the plasma up to the packaging of the final product each individual production step is controlled and documented.  Therefore, we provide our customers characterized with sera of consistently high quality, an absolute maximum level of safety and transparency.  In each batch of human serum among others the osmolarity, pH, protein- and glucose concentration, and cell growth are measured and the sterility tested.  Human Sera produced by PAN-Biotech consist exclusively of human material.  Human serum may be more suitable for the cultivation of human cells than FBS since serum and the cultured cells belong to the same species.  Thus, the physiological and natural conditions in vitro are reproduced most similar to those in vivo.  Human serum is particularly suitable for cell culture of human cells, tissue engineering of human tissue, human cell therapies (Clinical Trials I and II), particularly sensitive cell lines and immune cells.  PAN-Biotech offers three different categories of human serum: "Off the clot", Seraclot and converted human serum.  Within these three categories it is possible to choose between a mixed groups pool or human serum AB.

Biological Human

Adiponectin is a hormone exclusively expressed in adipose tissue. Many studies demonstrate that adiponectin has direct anti-diabetic, anti-atherogenic and anti-inflammatory functions. APM-1 can increase insulin sensitivity of skeletal muscle, attenuate hepatic lipogenesis and gluconeogenesis, regulate NO production in endothelial cells, inhibit proliferation of smooth muscle cells and prevent lipid accumulation of macrophage cells. In the circulation, adiponectin is present as three different oligomeric complexes, including the high molecular weight (HMW), the middle molecular weight (MMW, also called hexamer) and low molecular weight (LMW, also called trimer) forms. Different oligomeric complex of adiponectin activates different signaling pathways and exerts distinct functions. Adiponectin (Acrp30 Tri) human trimeric form was expressed in HEK293 cells. The Cysteine 39 was replaced with Alanine (C39A). Adiponectin C39A can only form a trimer, but not a hexamer or a HMW form. 

Biological Animal

GM-CSF is a monomeric glycoprotein that functions as a cytokine. It is a white blood cell growth factor. Granulocyte-macrophage colony stimulating factor (GM-CSF) is a hematopoietic growth and differentiation factor that stimulates the development of neutrophils and macrophages, and promotes the proliferation and development of early erythroid megakaryocytic and eosinophilic progenitor cells. It also stimulates growth in some epithelial cells and osteoclasts. GM-CSF is produced by a variety of cell types (monocytes, endothelial cells, T-cells, fibroblasts, mitogen-stimulated B-cells, and LPS-stimulated macrophages).

Antibiotic & Antibodies

Even though cells should be cultured without the addition of antibiotics or antifungal drugs, Cell culture media are not only a perfect breeding ground for your cell culture but also for many contaminants, such as bacteria and fungi. In certain cases the addition of antibiotics is necessary (for unique cell lines or to remove already existing contamination).  The OKT3 monoclonal antibody reacts with an epitope located on the ε chain of the CD3 complex. The human CD3 antigen is present on mature human T cells, thymocytes, and NKT cells. Clone OKT3 has been shown to activate T cell function.  In vitro, treatment of anti-CD3 stimulated T cells with PD-L1-Ig results in reduced T cell proliferation and IFN-γ secretion. Monoclonal antibodies targeting PD-1 that boost the immune system are being developed for the treatment of cancer.

Buffer Solution

The main purpose of buffered salt solutions is the maintenance of pH and osmolarity in cell culture applications as well as providing essential inorganic ions. These solutions are usually used for washing tissues and cells. Often they are combined with other agents in cell and tissue treatment.  Alsever's Solution is an isotonic balanced solution. It is used as an anticoagulant or blood preservative. Whole blood can be stored at refrigerator temperature up to 10 weeks.  In normal whole-blood erythrocytes are thousand times more frequently than leucocytes are and show an impediment for investigation or analysis of leucocytes. Erythrolyse buffers or Red-Cell-Lysing-Buffer is intended for the lysis of red blood cells in samples with white blood cells as for example EDTA-/citrate/heparin-treated whole-blood, Buffy Coats (PBMCs, leucocyte films), bone marrow. Red-Blood-Cell-Lysis-Buffer is specially used for lysing human erythrocytes in single-cell suspensions of peripheral blood and mouse hematopoietic tissues such as spleen. This buffer contains ammonium chloride, which lyses red cells, with minimal effect on lymphocytes when used as instructed. Nucleated red cells are not effectively lysed with ammonium chloride.

Amino Acid & vitamin

Amino acids are crucial for cell culture. Cells cannot synthesize essential amino acids and have to be added to cell culture While non-essential-amino acids relieve the intracellular metabolism and help cells grow better under the same circumstances. The availability of vitamins is extremely important for the cells, since the cells usually cannot synthesize the vitamins. Vitamins act as co-factors for many enzymes and are essential for their function. The absence of vitamins in culture may lead to decrease in cell growth, cell death or loss of productivity

Seperating Solutions

Seperating Solutions   Pancoll   In many cases the isolation of cells is the first step for gene expression studies or in diagnostic procedures.   Besides biological separation techniques physical separation methods are most commonly used.   These methods use physical differences such as size and weight of the particles to be separated.   For this purpose so-called separating solutions (= centrifugation media) are used.      Our Pancoll separating solutions contain a polysaccharide with a molecular weight of 400,000 daltons   This hydrophilic polymer allows for production of aqueous solutions for cell separation with a density of up to 1.2 g/ml.   PAN-Biotech offers a variety of ready-to-use products with a density of 1.063 g/ml up to 1.091 g/ml for a very wide range of cell separation applications

Serum Albumin BSA, HSA

Albumins serve as additive proteins for tissue cultures. They are the main protein component in serum and are added to cell culture media to increase the stability of cell membranes and to bind possibly toxic elements.  Human serum albumin is the most abundant protein in blood. Albumin transports hormones, fatty acids and other components. The main biological function is to regulate the colloidal osmotic pressure of blood. Human serum albumin e.g. can be used for protein calibration studies and solubilization of lipids.

Fetal Bovine serum FBS stem cell

Our specially developed, proprietary processing methodology for serum enables us to offer a special fetal bovine serum for embryonic stem cells (ES). PAN-Biotech's "FBS MSC" is especially tested for superior growth properties on human mesenchymal stem cells (hMSC). Cells are cultured in a medium containing 10% FBS of the tested batch. By flow cytometric analysis the cultured hMSCs are then assayed and analyzed for morphology, expansion and vitality of cells. Results are compared to those of a control serum which has shown good growth on hMSCs - and only the best batches are selected for "FBS MSC".

TEST KIT

Detection of mycoplasma-specific enzymes: A biochemical, highly sensitive and specific test kit is used to measure the activity of mycoplasma-specific enzymes. Living mycoplasmas are lysed and their enzymes are released. The released carbamate kinase and acetate kinase are incubated with the substrate mix. This leads to a conversion from ADP to ATP. The formed ATP in the sample is measured via a luciferase reaction. If mycoplasmas are present in the sample, a significant [increase] of the ATP level occurs, which leads to an intensified light reaction. Product recommendation: From a scientific and economic point of view, a sustained removal of mycoplasma from cell culture is absolutely essential. Not always can infected cell cultures be discarded and replaced with new ones. Infected cells also can not be treated with the standard antibiotics since the efficiency of this antibiotic treatment is usually insufficient. Mycorase 50x Solution (PAN-Biotech, Product number P06-02100) is a reagent for the permanent elimination of mycoplasma in cell culture.
